摘要(Abstract):

实时荧光定量PCR技术(real-time fluorescent quantitative PCR,FQ-PCR)以其特异性强、灵敏度高、重复性好、定量准确、自动化程度高、速度快、全封闭反应等优点在人类和动物疾病的快速检测、食品安全检测、定量分析、基因分型、基因表达研究、以及疫苗效力测定中成为分子生物学研究的重要工具,而且其定量范围极宽,无需做梯度稀释,特异性更强,克服了假阳性。本文分别从实时荧光定量PCR技术的背景、原理、类型、技术优点和定量方法、实验条件和主要影响因素、应用前景以及存在的不足等方面作了一个综述。
Real-time quantitative PCR becomes the important tool in the molecular biology research in human and animal diseases,food safety detection,quantitative analysis,gene expression,and determination of vaccine effectiveness.The background,principle,type,technology,the experimental conditions and the main influence factors and its application are reviewed.
